I m guessing that while dropbox doesn t edit the file the layer gets flattened with the visible layer and the resulting file is combined. If so it depends on other things like lighting sun direction if used and internal colour of the model you place it on as to how well the image rendors transparency. Place another layer beneath it and fill with colour.

First if there is a white layer in the backgroung where the layer is turned off in photoshop the white layer is still there but not visible. Png files tend not to respond well to being dragged and dropped but they behave like any other file once open in photoshop so i am thinking the the chequerboard is hardwired into the image and not actually transparent.
